What is the most comfortable, most durable and least amount of adjusting brand of these in a high-weight version? I rot in front of a desk 3/4 of my work day and need to do something.

I have been using a vest that I bought from Dick's Sporting Goods since around May. It will go up to 40# with 1# individual steel weights. Be careful with the vest and don't overdo it, I had to back off some because I started getting some lower back pain from overuse. 65-70# with a good back pack is much easier on the lower back than even 30-40# in a vest. It is adjustable and is about as comfortable as a weighted vest can be. I use mine 2X/week at 30# on a stair master for 30 minutes.
I also use ankle weights from Dicks Sporting Goods. Mine are 2.5# each and just strap around the ankle with a Velcro strap.

I have a weighted vest I got from Wal-Mart about 4 years ago, it maxes out at 20 pounds. I use it a lot for doing push-ups and going up and down the hills out by my house, when I am training it get used almost everyday. I can add more weight if I choose to. When I start training with heavier loads I just use my pack, its easier on the lower back and joints.
I have ankle weights but they never get used. I will do different leg exercise to strengthen my lower body. Squats, Lunges, Front Squats, Leg Press, Leg curls, Stiff Legged Dead Lift, Things like that and a lot of biking
