There might be several ways, but here's one. I started a new to me regime from the pages of Uphill Athlete- a muscular endurance program. Basically take a steep slope, load up roughly 25% of your body weight, put it in a pack and climb the slope (while keeping your heart rate in Zone 2—relatively low). I found a suitable that slope climbs ~1400' in 3/4 of a mile, loaded the pack w/ 45# and gave it a go. I finished off the hike by making it a 5 mile loop (after the 1400' climb).
I should also mention I haven't used my hunting boots or pack since last season. Feet totally comfortable w/ no hot spots, despite the steep (and rocky) climb and pack totally comfortable as well. Can't ask much more than that
