Drew a rifle ram tag in S66 which I believe is the highest average elevation unit in the state. Lots of glassing above timberline in big drainages. Some of these drainages are 10 miles long, so I imagine there’s the potential to glass 5+ miles.

Planning on a DIY backpacking trip, so weight is a consideration. Currently own some Swaro 12x50 EL’s and a Meopta S2 82 spotter. Are the 12x50’s enough glass for this trip or should I be considering adding some 15x56’s?

You will be fine with 12’s and spotter. That’s already a lot of weight.
